Spectrophotometry is one of the most commonly used technique in pharmaceutical and biomedical analysis. It is employed in quantitative purposes and with certain limitations for characterization of drugs, impurities, metabolites and related substances. The technique basically involves measurement the amount of a substance in solution absorbed by ultraviolet (190-380 nm) or visible (380 - 800 nm) radiation. Spectral selectivity can be induced and/or enhanced by a number of chemicals or instrumental technique such as difference, higher-derivative and dual wavelength spectrophotometry. In derivative spectrophotometric the absorbance (A) of a sample is differentiated with respect to wavelength (?) to generate the first, second, or higher order derivatives. In the context of derivative spectrophotometric, the normal absorption spectrum is referred to as the fundamental, zero order or 0 D spectrum. In this book simple, rapid, non hazardous, accurate, non expensive and precise derivative spectrophotometric method was developed and optimized for analysis of Benzimidazoles (albendazole and mebendazole) and Nitroimidazoles (metronidazole, metronidazole benzoate, and tinidazole) drugs were studied in pharmaceutical formulations and compare with their official method either HPLC which is expensive or non aqueous titration which is hazardous and not accurate. The work included the study of the expected interference of pharmaceutical excipients, used in these formulations by choice of different brand formulations. Derivative spectrophotometric techniques were applied for analysis of these formulations, the derivative procedures were based on the linear relationship between the concentrations of pharmaceutical substances and the amplitude of derivative peaks. The quality control laboratories and the pharmaceutical industries can use these method for routine analysis. The derivative method can be apply in the assay of drug substance in different dosage forms, and in different analytical areas, for natural substance, commercial substances as well as in the clinical and biochemical analysis.
